Tournament director Craig Tiley said unvaccinated players would not be allowed to compete at the 2022 Australian Open.
Defending men’s champion Novak Djokovic said he does not want to reveal his vaccination status publicly.
“Novak knew he would have to be vaccinated to compete,” Tiley said.
“We’d love to have him here.”
Tiley also confirmed that the tournament, which runs from January 17 to 30 in Melbourne, will be held in front of a large capacity audience.
Djokovic, 34, could claim his 21st major men’s title if he plays at Melbourne Park, where he is a nine-time champion.
Last month, the Serb said he wasn’t sure if he would play, adding he was waiting for confirmation from Tiley before making a decision.
Speaking earlier this week at the season-ending ATP Tour final in Turin, Djokovic said: “Freedom of choice is essential for everyone, not just me or anyone else, it’s not really important.
“Whether it’s vaccinations or anything else in life, you should have the freedom to choose, to decide what you want to do in this particular case, what you want to put in your body. “
The latest figures show that 80 of the top 100 men’s tennis players are now vaccinated against Covid-19. The Women’s Tennis Association (WTA) has yet to provide BBC Sport with vaccination rates among its players.

The British men’s cricket team, who are currently in Australia for the Ashes, spent two weeks in quarantine at a Gold Coast resort before they could begin full preparations.
